To directly probe the conformational equilibrium of the activation loop of mitogen-activated protein kinase p38α, we present an approach based on site-directed spin labeling, electron paramagnetic resonance (EPR) distance restraints, and multilateration. We demonstrate that the activation loop of apo p38α resides in a highly flexible equilibrium state and we reveal that binding of small molecules significantly alters this equilibrium and the populated sub-states.</dcterms:abstract> <dc:date rdf:datatype="http://www.w3.org/2001/XMLSchema#dateTime">2020-09-21T12:25:56Z</dc:date> <dcterms:isPartOf rdf:resource="https://kops.uni-konstanz.de/rdf/resource/123456789/29"/> <dspace:hasBitstream rdf:resource="https://kops.uni-konstanz.de/bitstream/123456789/50925/1/Roser_2-tskeesl8c0w13.pdf"/> <foaf:homepage rdf:resource="http://localhost:8080/jspui"/> <dcterms:title>Conformational selection vs. induced fit : insights into the binding mechanisms of p38α MAP Kinase inhibitors</dcterms:title> <dc:contributor>Drescher, Malte</dc:contributor> <dc:creator>Roser, Patrick</dc:creator> <dspace:isPartOfCollection rdf:resource="https://kops.uni-konstanz.de/rdf/resource/123456789/29"/> <void:sparqlEndpoint rdf:resource="http://localhost/fuseki/dspace/sparql"/> <dc:contributor>Weisner, Jörn</dc:contributor> <dc:contributor>Roser, Patrick</dc:contributor> <dcterms:hasPart rdf:resource="https://kops.uni-konstanz.de/bitstream/123456789/50925/1/Roser_2-tskeesl8c0w13.pdf"/> <dc:creator>Stehle, Juliane</dc:creator> <dc:contributor>Rauh, Daniel</dc:contributor> </rdf:Description> </rdf:RDF>
