Here, we showed that different inflammatory stimuli provoked oligomerization of the chemokine receptor CCR7, enabling human dendritic cells and T cell subpopulations to process guidance cues not only through classical G protein-dependent signaling but also by integrating an oligomer-dependent Src kinase signaling pathway. Efficient CCR7-driven migration depends on a hydrophobic oligomerization interface near the conserved NPXXY motif of G protein-coupled receptors as shown by mutagenesis screen and a CCR7-SNP demonstrating super-oligomer characteristics leading to enhanced Src activity and superior chemotaxis. Furthermore, Src phosphorylates oligomeric CCR7, thereby creating a docking site for SH2-domain-bearing signaling molecules. Finally, we identified CCL21-biased signaling that involved the phosphatase SHP2 to control efficient cell migration. Collectively, our data showed that CCR7 oligomers serve as molecular hubs regulating distinct signaling pathways.</dcterms:abstract> <dc:creator>Hauck, Christof R.</dc:creator> <dc:creator>Krueger, Wolfgang A.</dc:creator> <dc:contributor>Hauck, Christof R.</dc:contributor> <dspace:isPartOfCollection rdf:resource="https://kops.uni-konstanz.de/rdf/resource/123456789/28"/> <dc:contributor>Krueger, Wolfgang A.</dc:contributor> <dcterms:hasPart rdf:resource="https://kops.uni-konstanz.de/bitstream/123456789/33063/1/Hauser_0-321045.pdf"/> <dc:creator>Boyman, Onur</dc:creator> <foaf:homepage rdf:resource="http://localhost:8080/jspui"/> <dc:creator>Hauser, Mark A.</dc:creator> <dc:date rdf:datatype="http://www.w3.org/2001/XMLSchema#dateTime">2016-02-19T13:53:51Z</dc:date> <dc:creator>Schaeuble, Karin</dc:creator> <dcterms:issued>2016</dcterms:issued> <dc:contributor>Impellizzieri, Daniela</dc:contributor> <dc:contributor>Schaeuble, Karin</dc:contributor> <void:sparqlEndpoint rdf:resource="http://localhost/fuseki/dspace/sparql"/> <dcterms:isPartOf rdf:resource="https://kops.uni-konstanz.de/rdf/resource/123456789/28"/> <bibo:uri rdf:resource="https://kops.uni-konstanz.de/handle/123456789/33063"/> <dcterms:available rdf:datatype="http://www.w3.org/2001/XMLSchema#dateTime">2016-02-19T13:53:51Z</dcterms:available> <dc:contributor>Boyman, Onur</dc:contributor> <dc:contributor>Kindinger, Ilona</dc:contributor> <dcterms:title>Inflammation-Induced CCR7 Oligomers Form Scaffolds to Integrate Distinct Signaling Pathways for Efficient Cell Migration</dcterms:title> <dc:creator>Impellizzieri, Daniela</dc:creator> <dc:creator>Kindinger, Ilona</dc:creator> <dspace:hasBitstream rdf:resource="https://kops.uni-konstanz.de/bitstream/123456789/33063/1/Hauser_0-321045.pdf"/> </rdf:Description> </rdf:RDF>
