We investigated in mice immunosuppressed by different immunosuppressives (cyclosporin A, dexamethasone, tacrolimus and sirolimus), whether the innate immune system can be reactivated by granulocyte-macrophage colony stimulating factor (GM-CSF) or interferon-gamma (IFNg). Both study drugs reconstituted the suppressed macrophage response ex vivo and in vivo, while the IL-2 and IFNg response of T-cells remained silenced. Mice, immunosuppressed by cyclosporin A or dexamethasone survived a lethal Salmonella typhimurium infection when pretreated with GM-CSF or IFNg without inducing skin graft rejection after allotransplantation.
