We found that the efficiency of cross-priming in mice after vaccination with biodegradable poly(D,L-lactide-co-glycolide) microspheres (MSs) was enhanced when ovalbumin was coencapsulated together with either a CpG oligonucleotide or polyI:C as compared to co-inoculation of ovalbumin-bearing MS with soluble or separately encapsulated adjuvants. A single immunization with MS containing coencaspsulated CpG and ovalbumin yielded 9% SIINFEKL/H-2K(b) tetramer positive CTLs, production of IFN-gamma, efficient cytolysis, and protection from vaccinia virus infection. Taken together, coencapsulation of adjuvant and antigen is an important paradigm for the generation of potent CTL responses.</dcterms:abstract> <dc:creator>Basta, Sameh</dc:creator> <dc:rights>deposit-license</dc:rights> <dc:creator>Groettrup, Marcus</dc:creator> <dc:contributor>Fischer, Stefan</dc:contributor> <dc:contributor>Busch, Dirk H.</dc:contributor> <dc:creator>Müller, Marc</dc:creator> <dc:contributor>Müller, Marc</dc:contributor> <dc:language>eng</dc:language> <dc:creator>Fischer, Stefan</dc:creator> <dc:creator>Busch, Dirk H.</dc:creator> <dcterms:title>TLR ligands and antigen need to be coencapsulated into the same biodegradable microsphere for the generation of potent cytotoxic T lymphocyte responses</dcterms:title> <dc:date rdf:datatype="http://www.w3.org/2001/XMLSchema#dateTime">2011-03-23T09:06:45Z</dc:date> <bibo:uri rdf:resource="http://kops.uni-konstanz.de/handle/123456789/1194"/> <dc:contributor>Schlosser, Eva Christine</dc:contributor> <dc:contributor>Groettrup, Marcus</dc:contributor> <dcterms:issued>2008</dcterms:issued> <dcterms:bibliographicCitation>Publ. in: Vaccine 26 (2008), 13, pp. 1626-1637</dcterms:bibliographicCitation> <dc:contributor>Gander, Bruno</dc:contributor> <dc:creator>Gander, Bruno</dc:creator> <dcterms:available rdf:datatype="http://www.w3.org/2001/XMLSchema#dateTime">2011-03-23T09:06:45Z</dcterms:available> <dc:contributor>Basta, Sameh</dc:contributor> <dcterms:rights rdf:resource="http://nbn-resolving.org/urn:nbn:de:bsz:352-20140905103416863-3868037-7"/> <dc:creator>Schlosser, Eva Christine</dc:creator> </rdf:Description> </rdf:RDF>
